Title Inhibitory effects of geranium essential oil on the degranulation and cytokine production of mast cells

Abstract Objectives: Mast cells have critical roles in various biologic processes related to allergic diseases. The degranulation of mast cells occurs after the interaction of multivalent antigens with surface-bound Immunoglobulin (Ig) E. The secretion of many chemical mediators leads to the de novo synthesis of inflammatory cytokines as tumor necrosis factor (TNF)-alpha. In contrast, geranium essential oil (GEO) has been used for many years in traditional medicine as an anti-bacterial, antioxidant, and anti-inflammatory agent, but the roles of GEO in allergic reactions are incompletely understood. We investigated the effects of GEO on anaphylaxis.
Materials & Methods: We investigated the effects of GEO on the degranulation of cultured mast cells (CMCs) derived from mice to examine its anti-allergic activity. The degranulation of CMCs was determined using an assay to measure beta-hexosaminidase release. Cell viability was determined by applying the MTT assay. We also examined if GEO inhibited IgE-induced production of TNF-alpha. Levels of TNF-alpha in culture supernatants of CMCs were measured by a sandwich ELISA. Analyses of the chemical composition of GEO were undertaken by GC-MS.
Results & Findings: We found that GEO dose-dependently inhibited the degranulation of CMCs activated by IgE. Viability of CMCs did not decrease after incubation with GEO for 24h. Treatment with various concentrations of GEO before CMCs activation with IgE significantly inhibited the increased TNF-alpha level in a dose-dependent manner. Furthermore, citronellol (the major component of GEO) inhibited the degranulation and TNF-alpha production of CMCs.
Conclusion: These results suggest that GEO could be a candidate compound for the effective treatment of allergic diseases.
